MMP3 (Matrix Metalloproteinase 3) encodes stromelysin-1, an enzyme that degrades multiple components of the extracellular matrix including proteoglycans, fibronectin, laminin, and collagen types III, IV, and V. MMP3 also activates other MMPs including collagenases, making it a key regulator of matrix turnover and remodeling.The -1171 5A/6A promoter polymorphism in MMP3 affects gene expression, with the 5A allele associated with higher transcription. Higher MMP3 levels accelerate connective tissue breakdown and have been associated with skin aging, arterial aneurysms, and progressive joint destruction in arthritis. The 6A allele may protect against matrix degradation but could impair tissue remodeling.
