Overview

KLK4 (Kallikrein Related Peptidase 4) encodes a serine protease that plays essential roles in dental enamel maturation and is also expressed in the prostate. In tooth development, KLK4 degrades enamel matrix proteins during the maturation stage, allowing proper mineralization of enamel. This process is critical for producing hard, durable tooth enamel.

Mutations in KLK4 cause hypomaturation amelogenesis imperfecta, a condition characterized by soft, discolored enamel that chips easily. The teeth develop normally in shape but have defective enamel mineralization. KLK4 is also studied in prostate cancer research due to its expression pattern in prostatic tissue.
