PPARG (Peroxisome Proliferator Activated Receptor Gamma) encodes a master regulator of adipocyte differentiation and lipid storage. This nuclear receptor controls genes involved in fat cell development, insulin sensitivity, and glucose homeostasis. PPARG is the molecular target of thiazolidinedione drugs used to treat type 2 diabetes.The Pro12Ala (rs1801282) polymorphism affects PPARG activity and is one of the most replicated genetic associations with type 2 diabetes risk. The Ala allele reduces diabetes risk modestly but consistently across populations. This variant also influences body weight, insulin sensitivity, and response to dietary fat. PPARG activation improves insulin sensitivity but promotes weight gain through enhanced fat storage.
