NQO1 (NAD(P)H Quinone Dehydrogenase 1) encodes an enzyme that detoxifies quinones through two-electron reduction, preventing generation of reactive oxygen species. NQO1 is part of the cellular antioxidant defense system and is induced by oxidative stress through the Nrf2 pathway. The enzyme also stabilizes the tumor suppressor p53 and is important for vitamin K metabolism.The Pro187Ser (C609T, rs1800566) polymorphism results in a virtually non-functional enzyme when homozygous. The NQO1*2 allele is associated with reduced protection against oxidative stress, increased susceptibility to certain cancers, and altered response to chemotherapeutic agents that require NQO1 activation. The variant also affects coenzyme Q10 and vitamin K metabolism.
