IL33R (also known as IL1RL1 or ST2) encodes the receptor for interleukin-33, an alarmin cytokine released from damaged cells. The IL-33/ST2 pathway is a critical mediator of type 2 immune responses, playing important roles in allergic inflammation, asthma, and tissue repair. ST2 exists in both membrane-bound and soluble forms, with the soluble form acting as a decoy receptor.Variants in IL1RL1/ST2 are strongly associated with asthma susceptibility and severity, eosinophilic inflammation, and allergic diseases. The IL-33/ST2 axis is a major driver of allergic airway inflammation and is emerging as a therapeutic target for severe asthma. Soluble ST2 levels also serve as a biomarker for heart failure and cardiac stress.
