HLA-DPB1 (HLA Class II Histocompatibility Antigen DP Beta 1) encodes the beta chain of the HLA-DP heterodimer, an MHC class II molecule that presents extracellular peptide antigens to CD4+ helper T cells. HLA-DP molecules are expressed on antigen-presenting cells including dendritic cells, macrophages, and B cells. The gene is highly polymorphic, with hundreds of known alleles affecting immune responses.HLA-DPB1 matching is increasingly important in hematopoietic stem cell transplantation. Mismatches at this locus are associated with graft-versus-host disease, though the clinical significance depends on which specific alleles are mismatched. Additionally, HLA-DPB1 variants are associated with chronic beryllium disease in exposed workers and may influence susceptibility to certain autoimmune conditions.
