GSTP1 (Glutathione S-Transferase Pi 1) encodes a phase II detoxification enzyme that conjugates glutathione to various electrophilic compounds, facilitating their elimination from the body. GSTP1 is the most abundantly expressed GST in the lung, skin, and many other tissues, playing a key role in protecting cells from oxidative damage and xenobiotics.The common I105V polymorphism in GSTP1 affects enzyme activity and substrate specificity. This variant has been studied extensively for associations with cancer risk, particularly lung cancer in smokers, and with response to platinum-based chemotherapy. The variant enzyme has reduced activity toward some carcinogenic substrates, potentially affecting individual cancer susceptibility.
